中药学专业论文.doc目的 观察大黄对慢性肾功能不全大鼠肾功能及肾组织TGF-P 1和smad7表达的影响。 方法 采用腺卩票吟法建立大鼠慢性肾功能不全模型,治疗组予以大黄(4 g/kg/d)灌胃。治 疗4周后,检测各组大鼠的24小时尿蛋白定量、肾功能,通过组织学及蛋白印记观察肾组 织TGF-P 1和smad7的表达。结果 治疗组大鼠的24h尿蛋白定量、肾功能明显增加(P< 0.01),而治疗组大鼠24 h尿蛋白定量、肾功能较非治疗组减少(P<0.05)o非治疗组大鼠 肾组织TGF-Bl、smad7表达较正常组明显增加(PV0.01)。绕 大黄可降低慢性肾功能 不全大鼠24小时尿蛋白、肾功能水平,抑制肾组织TGF-B 1和smad7表达。大黄对慢性肾 功能不全的保护机制可能与降低肾脏TGF-P 1和smad7表达有关

ABSTRACT
Objective To study the effect of Dai Huang on glomerulosclerosis and expression of TGF-pi and smad7 in rats with CRF. Methods Rats were divided into control group, not treated group and Dai Huang treated group with 20 rats in each group. The rats in the latter two groups were intragastric of Adenine, the treated group was fed with Dai Huang at a dose of 4g/(kg-d) for 4 weeks. Urine protein excretion, urea nitrogen and creatinine was measured. The expression of TGF-pi and Smad7was measured. Results Compared with the control group, the not treated group showed higher urine protein excretion, urea nitrogen, creatinine(P<0.0 l)and expression of TGF-pi and smad7. Conclusion Dai Huang can diminish urine protein excretion and prevent glomerulosclerosis in subtotally nephrectomized rats. This protective effect is presumed to be associated with its effect on expression of TGF-pi and Smad7.
